    PG-013 is a fully-chemically synthesized, heterodimeric peptide with equivalent function to IL-15. PG-013 binds to IL-2Rβ and γc and activates downstream JAK/STAT5 signaling, similar to activities of recombinant IL-15. PG-013 can be used in many applications and protocols including scale up of CAR T and CAR NK cell therapies.

    PG-013 is anticipated to have comparable activity to recombinant human IL-15 when expressed on a ng/mL basis. To identify the optimal concentration under the specified test conditions, begin at the same concentration used for rhIL-15 and evaluate a dose series centered on that value (suggested points: 1/3×, 1×, and 3×).

    PG-013 has the potential to increase cytotoxicity of NKT cells.

    Frozen immature NKT cells were thawed and cultured for 10 days in medium containing either IL-15 or PG-013 to induce proliferation and maturation. The mature NKT cells were then evaluated for cell proliferation, and cytotoxicity activity against K562 cells. PG-013 induced robust cell proliferation (>12-fold increase) and yielded PG-013-matured NKT cells exhibited cytotoxic activity equivalent to or even exceeding that of IL-15-matured NKT cells.
