Product Basics
Thrombopoietin (TPO) stimulates the proliferation and maturation of megakaryocytes and is commonly used for the differentiation of iPSCs into hematopoietic stem cells and manufacturing of allogeneic cell therapy products, such as CAR–T cells.
PG-010 is an alternative to TPO that binds to the thrombopoietin receptor (TPOR) and functions as an epidermal growth factor substitute. It is confirmed to activate ERK in the TPO signaling pathway, as well as the JAK2-STAT5 signaling pathway, similar to TPO. It exhibits agonistic activity equivalent to recombinant TPO and shows comparable activity at 1/2.6 of the concentration of recombinant TPO due to a difference in molecular weight

Technical Information
(1) PG-010 demonstrated activity nearly equivalent to TPO
Recombinant TPO and PG-010 activities were compared using ERK phosphorylation and reporter gene activation under JAK2-STAT5 signaling pathway. The results indicated that PG-010 demonstrated activity nearly equivalent to TPO on a molar basis, showing similar activity at 2.6 times less concentrations than recombinant TPO based on mass concentration (ng/mL).
(2) Differentiation of iPSCs into NKT cells through hematopoietic stem and progenitor cells (HSPCs), using PG peptides (PG-007, 008 and 010) or using VEGF, CHIR99021 (Wnt3a agonist), and TPO.
PeptiGowth Inc. demonstrated the differentiation of iPSCs into NKT cells through differentiation into hematopoietic stem and progenitor cells (HSPCs) using PG-007, PG-008, and PG-010, and compared the productivity of HSPCs and NKT cells and the functionality of NKT cells with the control condition using VEGF, CHIR99021 (Wnt3a agonist), and TPO.
