PeptiGrowth PG-004
Noggin-like Peptide (BMP4,7 Inhibitor)
Product Basics
Noggin is commonly used in the culture of organoids derived from intestinal, pancreatic, lung, and tumor tissues, as well as for the neural and endoderm differentiation of stem cells.
PG-004 inhibits the BMP4 and BMP7 families in the same manner as Noggin, exhibiting antagonist activity against them. It exhibits comparable activity at 1/8 of the concentration of recombinant Noggin due to a difference in molecular weight.
Key Features
- Alternative to Noggin
- Inhibits the BMP4 and BMP7 families in the same manner as Noggin
- Comparable activity at 1/8 of the concentration of rNoggin*
- Stable. >90% remained after 14 days in serum free media at 37˚C
- Fully synthetic and animal-free
GMP grade can be manufactured upon request.
*Please note that this figure is an example and concentration optimization is needed.
Technical Information
(1) PG-004 exhibits the same level of inhibitory activity at 1/8 of the concentration (ng/mL) of recombinant Noggin.
PeptiGrowth Inc. conducted a functional comparison test between their Noggin-like Peptide (PG-004) and commercially available recombinant Noggin by measuring Smad1 phosphorylation. The results revealed that PG-004 exhibited inhibitory activity against BMP4 and BMP7 at the same level per molecule as recombinant Noggin. Furthermore, it was confirmed that PG-004 also demonstrates inhibitory activity against BMP family proteins with high sequence homology to BMP4 and BMP7. The molecular weight of PeptiGrowth’s Noggin-like Peptide is approximately 2.9 kDa, which is roughly 8 times smaller than the molecular weight of recombinant Noggin. PG-004 exhibits the same level of inhibitory activity at 1/8 of the concentration (ng/mL) of recombinant Noggin.
(2) Formation of lung alveolar organoids using lung progenitor cells differentiated from iPSCs using either PG-004 or Noggin (Day 14).
The induction of lung progenitor cells (LPCs) from hiPSCs and the formation of lung alveolar organoids using LPCs were demonstrated with PG-004. The concentration of Noggin was set at 100 ng/mL as a reference, while the concentration of PG-004 was varied within the range of 12–48 ng/mL.
- PG-004 (48ng/mL) yielded a comparable differentiation efficiency to LPCs to that of Noggin (100ng/mL).
- Lung alveolar organoids generated using PG-004 were found to be identical to those induced using Noggin.
